PFGE genotyping
Pulsed Field Gel Electrophoresis (PFGE) is method for separation of large DNA molecules on agarose gels using pulsed electric field. By this method it is possible to resolve complete chromosomes of bacteria and some eukaryotes (on PFGE is possible to resolve DNA fragments from few kb to few Mb in size). PFGE combined with digestion of total DNA (in agarose blocks) by restriction enzymes is used for genotyping of microorganisms and for detection of genomic rearrangements (deletions, duplications, translocations and inversions). In scientific literature PFGE is called “gold standard” because it is the most accurate method for the determination of the differences among strains of the same species. In recent years new rapid molecular methods for determination of microorganisms are developed, but PFGE still retains the position of the most accurate and most reliable methods, especially when one have to establish the differences/similarities between closely related microorganisms. In addition to determining the differences between strains PFGE is used for determination of relatedness among strains/species (dendrograms).
